Single-Cell Sorting of Oligodendrocytes and Oligodendrocyte Precursor Cells

A customer spotlight on Tim Czopka, Laura Hoodless and colleagues (University of Edinburgh) using FACSmax™ for single-cell sorting of oligodendrocytes and oligodendrocyte precursor cells (OPCs). Their research offers new insights into OPC heterogeneity, the regulation of oligodendrocyte lineage formation, and the responses of OPC subgroups to neural activity.

Tim Czopka, Laura Hoodless and colleagues (now at the Centre for Clinical Brain Sciences, University of Edinburgh, UK) used FACSmax™ from AMSBIO for Fluorescence Activated Cell Sorting to obtain single-cell oligodendrocytes and oligodendrocyte precursor cells (OPCs) from Zebrafish. (See Figure 1 for images of Transgenically labelled Oligodendrocytes following dissociation in FACSmax™).

Analysis of differentiation and proliferation of such single OPCs, published in Nature Neuroscience demonstrated the presence of two functionally distinct groupings of OPCs in the zebrafish spinal cord; providing new insights into OPC heterogeneity, the regulation of oligodendrocyte lineage formation, and the responses of OPC subgroups to neural activity.

Oligodendrocytes are highly specialized neural cells which form an insulating layer of myelination around axons in the Central Nervous System. Such myelin sheaths allow electrical signals to transmit quickly and efficiently along nerves – so that damage to the myelin can slow these impulses, as in Multiple sclerosis (MS) and other demyelinating diseases. We used FACSmax Cell Dissociation Solution from AMSBio to sort fluorescently labelled oligodendrocyte precursor cells from zebrafish embryos by fluorescence activated cell sorting, which allowed us to effectively isolate cells for single cell RNA sequencing. Using this approach, we were able to investigate oligodendrocyte heterogeneity by combining the molecular signature of oligodendrocyte precursor cells obtained from sequencing analysis with the cell fates of individual cells in vivo. This integrative approach revealed that oligodendrocyte lineage progression from early precursor to myelin forming cells is not linear for all cells, but that subsets of oligodendrocyte precursors exist that have different properties and fates.
